I am measuring the activities of catalase and SOD in my tissues homogeantes. I am wondering if we can measure superoxide levels in plasma or tissue lysates as it is converted to hydrogen peroxide by SOD1/2. If so, please let me know the technique.

You can still measure superoxide, as there is limit to how fast and effectively it can be dismutated to H2O2. Dihydroethidium (DHE) is probably the most commonly used probe for superoxide. It should be relatively simple to look up a published protocol that best fits your experimental set up.
