You need to determine the recovery of your analystes.

Secondly, if you will use an internal standard method and the calibration procedure will include the IS addition to the sample prior to sample preparation stage, the recovery factor should be satisfactory, but it don't have to be 100% - it must be just reproducible.

You can check this papers - the matrix is different, but the approach for recovery determ. and clibration the same as I suggested above.

Quatitatively add the ingredient, that you want to extract and measure, in different concentrations into food sample. Then you extract these food samples and analyze the concentrations of the ingredient in each of your extracted sample. If you extraction is efficient, the differences among the analyzed concentration will be very close to the differences among concentrations you added to your samples.
