The best way is through optical measurement using calcium indicators, either chemical or genetically encoded.
There are many indicators, some of which are semi-quantitative (detect changes and relative amplitudes) while others (radiometric) are quantitative.
You can find many studies on the topic, preferably studies using the cells of interest.
