how to measure the calcium level in cytoplasm of cells when stimulated by particular agonist for calcium channel?
Dear Yassir Zargar
The best way is through optical measurement using calcium indicators, either chemical or genetically encoded.
There are many indicators, some of which are semi-quantitative (detect changes and relative amplitudes) while others (radiometric) are quantitative.
You can find many studies on the topic, preferably studies using the cells of interest.
Good Luck
These are good protocols for Fura-2 calcium indicator (quantitative).
