We have a doxycyline-inducible adenovirus construct containing a probably toxic gene. The gene has no tags, nor are there any good antibodies against the protein.

To measure the transduction efficiency of the adenovirus, we would like to know if there are any methods to use without needing to express our protein of interest.

Is it possible to use an antibody and immunostain against the viral hexon protein to see the efficiency of the transduction, or is this just possible in virus-producing cells, i.e. cells expressing the E1 protein?
