I want to make Std mixture with IS C17:0 to analyze fatty acid composition of meat sample. I make Std mixture containing all FAs and IS separately (0.1g/10 ml hexane). But I don't understand in what ratio mixed both or separately run by GC. So, need cordial help in this regards.

Are you planning to do it as free acids or FAMEs or FAEEs?

OK for that you need a good separation from each FAMEs. Add C17 in nearly the same concentration that you will determinate the others FAMEs.
We prepare a mix of all FAtty acids + C17 acid
And to every sample we add the same quantity of C17 acid as the point in the calibration curve
That means that you add C17 in the quantity to each point in the calibration point, to each sample and the each QC sample.
Check first several samples that in your GC-FID conditions nothing overlap in Rt with the C17 FAMEs.
