• Use a specialized medium like RPMI 1640 supplemented with appropriate nutrients, such as 2 mM L-glutamine, 1% nonessential amino acids, 1% pyruvate, 50 μg/ml kanamycin, 0.05 mM 2-mercaptoethanol, and 10% Fetal Calf Serum (FCS). Supplement with cytokines like Flt3 ligand (Flt3L) to promote pDC survival and function. Check for viability before the assay.
