I don't understand your question. When I looked up T-agar medium on the internet, it comes up as Trichophyton Agar. This medium contains no yeast extract. It is used for growing the fungus Trichophyton, sp. Is this the medium that you are using? Please let me know and I can try to answer you.
I completely forgot to include the name of the agar medium. We intend to use the Tributyrin agar medium because we want to optimize the Lipase enzyme and see what happens if the yeast extract concentration is reduced. However, since the Tributyrin agar already contains yeast extract, we want to know how we can reduce it.
Attached is a document w/ the composition and usage directions for Tributyrin Agar (TBA). You may have already seen this.
If you want to reduce or modify the yeast extract (YE) concentration, the way to do it is to prepare the TBA "from scratch", i.e., don't use the prepared TBA. Instead, make it up yourself from the separate (individual) components. As supplied by the manufacturer, TBA contains 5 g/L of HiVeg peptone, 3 g/L YE and 15 g/L of agar. You can make up your own TBA w/ the required amounts of HiVeg peptone and agar, then add varying amounts of YE to prepare several batches of TBA, each w/ whatever YE concentrations you want, for example 0.5-, 1.0-, 1.5-, etc., g/L of YE.. It will take a little more time and effort, but it should work fine. I have done this before with other media where it was necessary to vary the concentration of one or more of the components.
If you don't have HiVeg peptone, you should be able to use another source, e.g., Bacto Peptone. Also, instead of 15 g/L of agar, you might want to use 20 g/L. I usually use 20 g/L agar. This will give you "stiffer" plates. This is not critical.....just a suggestion.
