I would like to use platelets in a co-culture with other cells.
I was wondering which are the conditions to keep alive the platelets:
-since I am going to buy my platelets, I was wondering if they can be shipped on ice
-which buffer do I need to keep them alive during the shipment?
-I saw some papers where they were kept in culture with RPMI, do you have any suggestion/ideas about that?

I hope you are very well. Regarding the platelets, it's depends on what is your aim so if you want to study the intracellular signaling pathways that is mean you need to fresh platelets and you can isolated by the washing platelet protocol or if you want to study the stabilization role so you can store it at -20 after isolated and then use it at a desired concentration in your further procedure.

Thank you for your answer. In my case, I would like to see if the platelets can be educated from the environment in which I am going to seed them. For this case, do you think I need to isolate them from plasma right before use?

I think yes you need to isolate human platelets from healthy blood donor first and then you can use it as fresh and live cells in the culture. Also, you can store some platelets in normal fridge for 24 h to see is the cold platelets (CP) is similar to the fresh platelets in terms of environment adaptation? This is my suggestion.

Therefore, I have to imagine that it is not possible to freeze the platelets (e.g. like we do with cells with DMSO) and then use them later. Am I right?
Sorry for delay in response. I think you can try that as a third condition (fresh platelet at RT, cold platelet and freeze platelets) and compare between these conditions. I used freeze platelet at -20 in the thrombosis stability before.
