Dear Mukesh - First, you have to make sure that you eliminate any microorganisms present on the insect cuticle by washing the insects in a 0.5% sodium hypochlorite solution (with gentle shaking for several minutes), followed by three rinses in sterile distilled water, and finally drying with sterile paper towels. After that, you can simply plate the "surface-sterilized" insects on PDA media. Of course it's recommended to add antibiotics such as tetracycline, streptomycin, penicillin, etc to the plating media for suppression of bacterial outgrowth.
Place the bodies under proper moisture conditions so that the growth of the fungus is carried out on their body surface. Then use a needle to picking up some of the fungus mass. Put it on the proper culture medium of your fungus.
Another way is to sterile dead insects and put them directly on the surface of the appropriate fungus medium.
Dear Mukesh, first of all you need to surface sterlized the the insects, this can be done either by 70% alcohol or 1-2% sodium hypocclorite. in either case you need to wash off the remaining chemical on the surface of insect cadavers by rinsing it three times ( alaglag as indian says) in sterlized distlled water . then get dry in again sterliced filter papers. of course all of this work must be carried out under aseptic conditions( laminar flow for example. instead of PDA you may use selective media if you are tatgeting certain EPF. this will save you time and effort. all the best
The paper in the link supplied by Yehia A.G. Mahmoud is very appropriate. However, carry out Koch Postulates after getting your isolates from the cotton whiteflies to ascertain the actual entomopathogenic isolate.
