I inoculated malt extract with spores from 5 day old malt agar plates. After that I incubated the flasks at 250 rom for 72 hr at 30C temperature. What I observed is the difference in fungal morphology. some flasks have pelleted growth while some have dispersed growth, but my product formation is in pellet growth. What will e the reason for this ? I am not filtering the spores, just scrape the spores from the plates with ( 0.9% saline solution and 0.01% triton X 100) solution, inoculate 100uL spores. Is this the reason or something else? Should I filter the spores and count it? Also let me know the factors favorable for pellet growth?

The best procedure for pellet formation in fungi is that it should be grown under static conditions. For getting maximum mass production of any fungi is that it should be grown in flat glass bottles having media in the slanting position.
Are you sure that you are working with a pure culture of Trichoderma?
