I am working on Trichoderma atroviride aromatic compound production. Earlier, I observed the spore as well as mycelial growth on the plate but now I am getting only spores on the plate without any mycelial growth. Due to this there is no product formation. However, the specific growth rate is same as compared to previous morphology. Then Why I am not getting any product. My main concern is product loss.

What type of media are you using? If your media is to dilute, then the fungi is struggling to survive, and will produce spores to survive another generation.
Are your cultures being incubated at the correct temperature? To hot of temperature could stress your mycelium and make them sporulate.
Are you incubating your cultures in the light or the dark, or combination of the two? Certain conditions will make your hyphae/mycelia produce spores.
There are a lot of reasons why you will not get hyphae/mycelium growth in a culture. I hope these help you troubleshoot your problem.

Hi, You can make mass multiplication of tricoderma on Jowar or barley seeds or rice bran which can be done by autoclave grains and some water should be added before inoculation of culture. after 15 days clear growth grain dried at room temp grind and can be use for fungal attack treatment.

For mycelial biomass, I suggest Potato-dextrose broth.
I think that the light stimulate sporulation and destro chemical métabolites.

Thanks for the response. I will try these methods and update accordingly. I am using Glucose 10g/L and 20 g/L malt extract as the plate media.
You can use PDA medium (Potato Dextrose Agar) with incubation at a temperature of 25 C, it's very rich medium and allow the fungus to produce large mass of hyphae. Also, you can use OA medium (Oat meal Agar) with incubation at a temperature of 20 C.
If you want to use PDA medium, you must prepare the medium by using 200 gm of potato, 20 gm of dextrose, 20 gm of agar and one liter of distilled water. They must autoclaved at 121 C and 15 pounds/inch2 for about 30 minutes. after that, you must add a suitable antibiotic to prevent the an wanted bacterial growth.
When you want to use OA medium, you must take 30 gm of oat meal, 15 gm of agar and one liter of distilled water. They mixed together carefully and autoclaved at 121 C and 15 pound/inch2 for 30 minutes, then you must add any antibiotic.
I think that pH below 6 (5 - 6), and temperature of 25 ْC can be good for mycelial growth on PDA.
