How to induce pyroptosis in THP1 cell?

In the experiment of inducing pyroptosis of THP1 cells, when adding ATP after pretreatment of cells with LPS, is it necessary to remove the LPS and then add the medium containing ATP, or add ATP directly to the medium containing LPS?

THP-1 cells were treated with 50 μM of each chemical in the bacterial chemical library for 1 h. After that, pyroptosis was induced with LPS stimulation at 1 μg/ml for 3 h followed by 10 μg/ml treatment of Nigericin for 1 h.
