I have to extract genomic DNA from skin cells collected on a swab. I am currently using the PureLink Genomic DNA Kit, and following the protocol for the Mammalian Cells Lysate, however they recommend to have at least 5 million cells, which I probably don't have by collecting skin swabs. I also thought about following their "Human Buccal Swab Lysate", however the cells collected in this case do not have the keratinocyte layer that skin cells have, so probably the digestion will be affected.
So far I am having very low 260/280, 260/230 values when I measure my sample on nanodrop.
Any ideas on which kit should I use or what should I avoid in my protocol?

I recommend using two pre-moistened swabs per sampling site to maximize your DNA yield.
Hello! They are pre-moistered! The problem is that the 260/230 values are too low when I do gDNA extraction.
