I need to increase the sensitivity of a paper based assay so that even trace amounts of antigen immobilized on nitrocellulose membrane would be able to capture the gold nanoparticle-antibody conjugate from the flow and show colour.
How do I increase the loading of nanoparticle-antibody conjugate onto the glass fiber conjugate pad ? As of now, I dip the conjugate pad in the nanoparticle-antibody dispersion. Only part of the conjugate is getting loaded onto the the conjugate pad as it gets completely filled up. How do I ensure that I load the maximum possible so that sensitivity can be increased.
Can anyone share their views ?

Put an assembly where you get a flow, steady and repeated (separate with little time gap), for loading since dipping is causing problem.
