I was trying to do AuNP based Lateral Flow Immuno Assay LFIA to detect Virus. I have 3 different antibodies for this virus. I was trying to do a sandwich LFIA using an antibody pair from these 3 Abs. I have tried with all 3 possible combinations of antibody pairs from these 3 Ab. I even switched the positions of the Abs on the conjugate pad and the test line. But none of these has given color at the test line, while control line is giving color in all the combinations.

Then, i wanted to see the affinity / binding of each of these 3 Abs individually towards the virus. I made AuNP-Ab conjugate with all the 3 Abs individually and coated them separately on 3 different conjugate pads and thus made 3 different strips having different Abs on the conjugate pad. Later, I coated virus at the test line and respective secondary IgG as the control line on these 3 strips. I ran all the 3 strips with 1X PBS. But none of them gave color at the test line while the control line is showing color. This means that the neither of the 3 AuNP-Abs is conjugating with the virus.

What could be the possible reason for this ?

The size of the virus is 40 nm in diameter and I am using the whole virus with the envelop.

The antibodies were tested positive for the virus using immunohistochemistry and immunofluorescence.
