I have been maintaining a line of N2As for a couple years in my current lab. In the last 4 months I have been having a lot of trouble keeping them growing. They are growing really slowly, and not surviving passages. Even previously frozen stocks have trouble thriving after 1 or 2 passages. We use DMEM, 10% FBS, L-Glu, Pen/Strep. My U2OS cells and 3T3 are fine.
Anyone have any suggestions for troubleshooting this problem?
