Is it possible to presume about which Salomonella species present in the sample without using molecular tools like PCR? Just from biochemical tests?
If I plan to do PCR for confirming species, how do I decide which primers I need to use? Studying literature for prevalent species information? Or there is any practical methodology?

You can use differents approaches:
- Novel culture techniques as: immunomagnetic separation, EIA- and ELISA-based assays incorporating fluorescent or colorimetric detection, simple lateral flow assays incorporating immunochromatographic technology
-The ISO-6579 standard (ref. 1) which recommends using the TSI agar, Urea agar (Christensen), L-lysine decarboxylase, β-galactosidase (ONPG), Voges Proskauer and Indole tests in this order.
-The LOUIS test is a screening protocol for Salmonella and Shigella that uses rapid enzyme test.
