Hello,

I am isolating primary murine glia cells from different neonatal knockout strains (P0-P4). I have issues getting single astrocyte cells after adding 4 ml of trypsin (2,5 %) to the PDL coated 75 cm² flasks (containing 2 brains). After resuspending the cells several times, they are still sticking together forming big clumps of cells (I am assuming astrocytes). I am resuspending the cells in DMEM medium using a 10 ml pipette. Since I need single cells for cells sorting via MACs, I am seeking for a method to get rid of all the cell clumps. Does anyone have experience with that problem or know any methods to prevent astrocyte cell clumping?
