I'm currently trying to perform DNA extraction from samples of scolopendromorph genus Newportia, but I don't know which is the best way to get muscular tissue from the samples. I read on most articles that the legs, parts of the body (like a segment, I guess) or even the entire body is used, however, I'm not sure if I should use the entire legs as dissected from the body or if I should cut it into small pieces, as well as I wonder if a segment from the body or the whole body will allow me to get good samples of DNA without contamination (and if that's the case, how's the procedure for extraction?). What would you recommend?
