In some preliminary experiments on a novel cell line, we have found that after inward current a secondary one is visible and we excluded it to be Ca++ wave and or an Na wave. The IV of this wave shows a reverse potential close to 0mV which is also our equlibrium potantial for Cl. This rises the hypothesis that can be a Cl dependent wave. So far, before to look at expensive pharmacological investigations, we would like to determine if this is a cloride inward current. The easiest way would be to remove both internal and external cloride and see if the wave disappear. Has anybody a good suggestion for solutions (both internal and medium) cloride free? Any suggestion or comment is welcome :)
Dear Jai, thank you very much for your answer, but probably my question was not clear. I was not worrying about residual Cloride in water. I was asking suggestions for good intra and medium solutions to prepare Without Cloride. For example, using K-Gluconate inside the micropipette is a start, but in our usual solutions we have also Calcium cloride and Magnesium cloride. Removing of Calcium make difficult the seal. So What can we use instead of CaCl2 and MgCl2? The same problem is for medium.
