In some preliminary experiments on a novel cell line, we have found that after inward current a secondary one is visible and we excluded it to be Ca++ wave and or an Na wave. The IV of this wave shows a reverse potential close to 0mV which is also our equlibrium potantial for Cl. This rises the hypothesis that can be a Cl dependent wave. So far, before to look at expensive pharmacological investigations, we would like to determine if this is a cloride inward current. The easiest way would be to remove both internal and external cloride and see if the wave disappear. Has anybody a good suggestion for solutions (both internal and medium) cloride free? Any suggestion or comment is welcome :)
