I have a few questions about cell smear fixing and storing.
1. what grade for the acetone did you use for fixation cell smear?
2. is the ACS reagent grade (≥99.5%) ok for fixing cell?
3. Did you wash the cell slides in 1xPBS or other washing buffer after fixing it in cold acetone and before storing them at -80C or 4C?
4.Is it needed to rehydrate the cell slides stored at -80C or 4C when you stained it?
5. Did you do antigen retrieval for acetone fixed cell smear slides?
6. Is it needed to treat the cell smear slides with 10% neutral buffered formalin (NBF) after fixed in acetone?
7. Is there a step to stabilize the cell smear on slides? if yes, how to do?
