besides being more than happy if you get an immune response at all, you could run tests with other polysaccharides that share parts of their structure with your molecule in order to narrow down the relevant subsctructures..
If you should go for hybridomas/monoclonals, you may sequence the clones, deduct the protein sequence of the paratopes (CDS/Fv) and perform in silico docking experiments with suspected candidate structures.
May I ask how you are coupling the polysaccharide to the carrier protein (which?) and what is the polysaccharide you're working with?
I am working with Brucella polysaccharide. I was able to produce a strong immune response with purified surface polysaccharide in comparison to conjugate.
