Hi. In the past I used glass pipettes to do stereotaxic injections and filled them via capillary action and used mineral oil to avoid dead space. Now in the lab we have WPI nanosyringes that are almost the same as Hamilton syringes. Problem with these syringes is that I cannot prime the entire 10 uL syringe with my AAV. we cannot waste that much AAV everytime we fill the syringe. Also, I am planning to inject only 200 nL per mice, so for a day of surgery with 4 mice I will only need around 1 uL of AAV. I am trying to figure out how to fill the syringes with such little volume.

Front filling using the bendy fiber needle that WPI provide is not going to work because a volume like 5 uL completely gets lost in the needle itself and when I push it in the syringe it itroduces lot of air and bubbles.

I think the solution has to be priming with some other solvent first to avoid bubbles and dead space but I am not sure what solvent to use. I certainly cannot use mineral oil because these nanosyringes are designed to overcome the problem in the first place why mineral oils are used. I am not sure if I can use ethylene oxide or methylene oxide to prime the syringe and the needle with AAV's. I would appreciate any solutions to this problem. Thanks!
