It depends on the nature of tissues and proteins to be extracted. There is no perfect method for all proteins, but the principal thing you must take in consideration: the choosen method shouldn't destroy the cell membrane(s).
The simplest method by dissolving extracellular proteins in adequat solvent from the tissues (regarding your sample), then precipitate proteins according to pHi or other separating techniques.
