Hello, I am trying to know tomato virus titer/concentration in samples and I will perform serial dilution of the positive control. I wanted to ask How to estimate/know plant virus concentration to make a standard curve in ELISA after serial dilution.
Hello, after serial dilution you can perform the calculation of linearity, repeatability, precision, but if you do not have the sample with know number of tomato virus inside of reference material you can not perform any quantification. If your positive control not providing any informaton about the quantity it will give you only qualitative data about existence of virus but not quantity.

dr. Karen A. Darbinyan is right, it is not possible to perform such analysis without knowing the quantity in your reference material.
If you would have the possibility to check your positive control in PCR you could determine the concentration comparing the intensity of your band with the marker and lambda DNA. An alternative is, of course, the Real Time PCR.
