You will need to measure the concentration of xylose (and maybe other hemicellulose sugars) in the liquid hydrolysate via HPLC. Then you can calculate back the percentage of xylan (and equivalent polymers for the other sugars), which add up to the hemicellulose. First determine the holocellulose then from it determine ⍺-Cellulose. Finally ⍺-Cellulose is deducted from holocellulose and you will get hemicellulose. You can follow the TAPPI method.
