Hello, I am trying to use siRNA to K/D gene in human PBMC cells. I was wondering which transfection reagent and protocol is the best for this. I am using RNAiMAX (like I did for human fibroblasts) for 24-36-48hrs without efficient transfection.
Just try the Amaxa system, nucleofection !

Indeed Amaxa system is the best approach,, or using transduction system by lentivirus.
PBMC are not easy to transfect by other way.

Just try the Viromer BLUE and GREEN! They work very good for most of mononuclear phagocyte cell lines. More than 90% silencing has been obtained on PBMCs.
