We have to carry out in vitro hydrolysis of lipophilic prodrug (log P 3.39) to drug in the presence of enzymes. As the drug is having very poor solubility in aqueous phase (phosphate buffer pH 7.4), we have to dissolve the prodrug in organic solvents (chloroform and methanol).

a) Is the activity of enzymes will be affected by organic solvents or not?

b) Even if we spike 100-200 uL of prodrug solution to aqueous phase, there is immediate precipitation of prodrug.

Kindly resolve our issue.
