"For the migration assay, the transfected cells (5 × 104 cells) were seeded in the upper chamber in 200 µl serum-free medium and 600 µl medium containing 10% FBS was added to the lower chamber with or without TGF-β1 (5 ng/ml). After 24 h, the cells were fixed with 4% polyformaldehyde (Solarbio, Beijing, China) for 15 min at room temperature and washed with PBS thrice. 0.1% crystal violet (Solarbio) was used to stain cells for 30 mins at room temperature and later was washed with PBS 3 times. Residual crystal violet was removed by gentle wiping using cotton bar. Inverted microscopy (Leica DMI4000 B) was used to capture migrated cells and the number of migrated cells were measured by ImageJ."
