I have 500 mg of Temozolomide powder. So, how to dissolve TMZ properly for stock solution (10 mg/ml)? Can I dissolve it in distilled water or PBS instead of DMSO? I will use this drug and load it into extracellular vesicles (exosomes) of U87 cells.
For cell culture experiments, TMZ is routinely dissolved in DMSO. It is also soluble in dimethyl formamide. Further dilutions of the stock solution may be made into culture media or aqueous buffers prior to performing biological experiments.
The solubility of TMZ in DMSO varies between 9.7 mg/mL and 39 mg/mL, corresponding to a stock concentration of 50–200 mM. TMZ is slightly soluble in water while its solubility in PBS is 0.33mg/ml.
You may make a stock solution of 10mg/ml in DMSO, and the working solution may be made in culture media or sterile PBS. Please note that the final concentration of DMSO in culture should be less than 0.1% as it is safe for the cells.

So, how to prepare 50 µl of these concentrations (10 µM, 50 µM, and 100 µM) with 0.1% DMSO? I'm still confused about how to make 0.1% DMSO final concentration using the drug to be used for the MTT assay.
