I'm facing a problem in dissolving two synthetic peptides (21 amino acids long). The peptides have iso-electric points at pH 4.01 and good predicted solubility in water. However, the synthesized peptide is not soluble in water, PBS, DMSO or acetonitrile. What might be the reason and kindly suggest some solution.

I've had similiar. If you can, try to add some aminoacid residues that would promote solubility on either (N or C or both) sides of the peptides and order a new peptide.
Since your peptide is acidic, it should be solubule in basic buffers. Maybe just try adding some NaOH.
Another interesting solution is to prepare 1 M arginine and set the pH level to the desired. It has been found that buffers or water containing 0.2-0.5 M arginine would promote solubility. Maybe try preparing different pH arginine solutions and try with that.
