Hi,
I would like to know how we can digest mouse tissue to measure ATP content via HPLC or some other plate based assay.
The assay kit we currently have tells us to homogenise the tissue but does that not release more ATP? I expect that the difference we are trying to detect to be small hence wanting to stay away from mechanical dissociation if possible. I've tried using TCA which interfers with the chemicals for the plate based assay.

I would first make a protocol on sample preparation that grantee to remove matrix effect
