I have a glial cell line called MO3.13. This is an immature cell line. I wan to differentiate into mature oligodendrocytes for my in vitro study? My question is how I can differentiate? What types of chemicals I have to use? Thanks
In order to differentiate MO3.13 cell line into mature oligodendrocytes, you just need to replace FBS from growth media by 100 nM PMA. This protocol generally takes 5-7 days to get maximum expression of mature oligodendrocytes marker such as MBP, MOG etc.
Growth media: DMEM High glucose + 10%FBS (Glutamax and Pen-Strep)
Differentiation induction media: DMEM High Glucose + 100nM PMA (Glutamax and Pen-Strep)
