Our TALEN nucleotide sequence is 3750 (ORF) and according to this our protein size should be approxinmately 137kda but size is coming around 75-100 kda. Any suggestions?

I assume you are estimating mass by size exclusion chromatography - this is shape as well as mass dependent. Mass can be determined by multi-angle laser light scattering if you have access to the equipment. Otherwise you can do mail-in SAXS of your sample and a matching buffer to get mass (see Nature 496, 477-81, 2013) and http://bl1231.als.lbl.gov. Of course you can also do sedimentation equilibrium with an analytical ultracentrifuge - this takes several days and again you need access to the equipment.
