I am trying to determine the ploidy level of bacteriods (Sinorhizobium meliloti from the root nodules) taking the free-living bacteria as the reference. I would like to know how to determine the ploidy of any polyploid organism with respect to the reference organism (known ploidy)?Is there anyone who can help me?

BD Biosciences provides a kit which can be utilised to determine the ploidy of cells, however it ultimately depends on your flow cytometry setup (i.e.: what flow cytometer, lasers and reagents are at your disposal etc).
This is the kit which will allow you to analyse DNA content / Ploidy. I would suggest you read through the protocols, reagents and see if it is applicable to what you aim to achieve.
