I prepared 1:1 inclusion complex of flavonoid and HP-b-CD through freeze drying method .
( obtained AL type graph through phase solubility study conducted with different concentrations of CD) . While finding the drug content in Inclusion complex, (1) 5 mg of pure drug and (2) 5 mg of IC (prepared by adding 27.2 mg of HP-b-CD and 6.64 mg of drug(1:1 molar ratio) via freeze drying method) was dissolved separately in water ,stirred on a magnetic stirrer for 24 hrs at RT. The resulting dispersion was then centrifuged at 10,000 rpm (5 min) and the supernatant was collected and filtered using 0.45 um syringe filter and the sample was analyzed using HPLC.
In case (1) where pure drug was used , HPLC showed strong peak at designated wavelength. But in Case (2) HPLC didn't not show peak at same position (or same retention time).
Do i need to add more IC (5 mg was taken) to evaluate the drug content in IC?
