Now one of my research projects is to couple the antibody with Carboxyl magnetic nanospheres to prepare the immunomagnetic beads. The coupling experiment has been accomplished according to the product protocol, but I have no ideal how to detect whether the antibody has successfully coupled with the carboxyl magnetic nanospheres and how to calculate the coupling efficiency. How to deal with the though question? Do the every member of the researchgate have a ideal method to resolve the problem?

I myself was looking for the answer but what I will suggest is to save an aliquot of antibodies before coupling reaction and get the antibody solution after the reaction and check out the concentrations of both using nanodrop. This will give you percentage efficiency of the antibodies, hope it works
Hey ! I also used the nanodrop to assess my coupling efficiency but I encountered interference problems (like the OD of my surnatant was much higher than my starting solution). Might come from trace interferences from the beads. I use LC size exclusion chromatography to detect presence of remaining Ab in my surnatant. But of course this requires your lab to be equipped with such instrument.
