Hi, I am working on optical trapping of silica particles of size 1-3 um dia. My calibration curve shows that the spring constant increases with power within this diameter range. The wavelength of laser used is 830 nm. I am using power within the range 200-440 mW.

It’s a bright field microscopy. I tried trapping it in water. Even though the spring constant comes out as increasing with power and the displacement curve of the particle shows that it is trapped, I am failing to obtain the correct viscosity value for the bead trapped in water.

What can be the possible reasons why my viscosity of water is not coming out correctly?
