Kindly , I have human ASCs and BM-MSCs cell lines from ATCC. They mentioned on the website that they are cultured on mesenchymal stem cells media ! However, I have on my lab DMEM low glucose media ? does it make a difference ? and they mentioned to be culture with bfgf and EGF? So, DMEM + 10 % FBS + L-Glutamine + Penicillin is not enough ?
DMEM low glucose (containing L-glutamine) + 10% FBS + 1% antibiotics (mostly penicillin-streptomycin) is appropriate for MSC culture. So, the media that you use should be okay for growing MSCs. Good luck!
FBS can be appropriately reduced, but the specific reduction needs to be explored.
I agree with Intan. However, they will send you passage (1-2?) cells that were growing with their medium and they might not be happy if you change the culture conditions! Also, I found bFGF essential for the growth of human ADSCs.
For BM-MSCs isolated from patients I usually use a medium of DMEM low glucose, 10% FBS, 1% of penicillin/streptomycin and 1% of L-glut, without any growth factors, while i prefer to increase FBS at 20% in the Adipo's medium, with rest of component at the same concentration.
