How to create stable cell line expressing big protein?

I want to create a stable cell line expressing a big protein (~6500bp).

I have approx. 12kbp between the 3-5' LTR seq of a lentivirus expression plasmid.

Has anyone succeeded doing so with such big protein in lenti expression system?

You would probably get a low yield of viruses but for a stable line you don't need more than a few infected cells so you should be fine.

Alternatively you can try switching to piggyBac which has a much larger size tolerance.
