If we have to correlate a soil parameter (say concentration of compound/ or N content/ enzyme activity/ or any function) analyzed by physico-chemical method and have data of bacterial diversity assessed by metagenomic approach, then how it may be correlated to reach a conclusion?

Typically, to find out whether or not the measured soil parameters in this case (soil physicochemical parameters) impact the microbial responses, we generally try to infer some sort of association between the two. For doing so we can use the diversity indices as a collective measure of the whole community. So for "n" samples being compared we can do Pearson's correlation coefficient measurement to check for observable trend (basically by regression modelling). To obtain better resolution and get a meaningful insight this can be iterated at different taxonomic level, if the full dataset fails to provide any meaningful insight. Similarly, when comparing the corresponding matrices Mantel's test can be done to find significant correlation (positive or negative) if any. The same set of tests were also carried out to study patterns of geographic variation in taxonomic data but recently Legendre et al., 2015 provide further insight in these type of spatial analysis.
