I need conserve my conidia in solution to use frequently without these germinate. At this moment I am using saline solution (NaCl 0.8%), but the conidia are germinating a few days after transfer in this solution.
You can conserve conidia in liquid nitrogen, PDA and mineral oil. The oil overlay technique is commonly used to conserve conidia for many years. Heavy grade mineral oil can be sterilized by autoclaving for 15 min at 121 °C and 15 psi pressure in half-filled 250-ml flasks. Fungal cultures can be incubated at 25 ° C for 2 weeks and grown on PDA screw-cap glass test tube slants. The fungal cultures is then covered with mineral oil up to 1 cm above the top of the slants, completely submerged in the oil. The tubes are stored in an upright position at room temperature.
