I'm working with some hydrogel carrier systems, which carry 166Ho-based nanoparticles, aiming at brachytherapy applications. I want to perform some in vitro tests with radioactive systems to optimize my compositions.
However, I'm struggling with the methodology. I do not know whether the methodology I'm planning to use is the most appropriate for this purpose. In brief, I'm planning to seed cells in culture wells of a 24-well plate; then, I'm going to insert a transwell containing my hydrogels containing 166Ho in the culture plate. Later, the cell will be incubated for 48 h. Finally, I'll perform an MTT assay to evaluate the cell viability. (Please, see the attached paper in the "In vitro brachytherapy" section within section 4. Experimental Section).
If anyone has a better methodology or can suggest a specific standard to follow, I would like to hear some advice.
