How to choose the size of FITC-dextran to determine mesothelial cell monolayer permeability?

I have to check the permeability of the mesothelial monolayer. I am using a 96-transwell plate with 8 μm pores, coated with type I collagen and then culturing the cells to form tight junctions and create a monolayer.

I am wondering what size FITC-dextran I should use for the assay?

I look forward to any suggestions.

Hi Veronika,

To evaluate the paracellular permeability, I would recommend to start from 4 kDa (and 10 kDa). If you are interested in size selectivity, you can also test fluorescein alone (~332 Da), 70 kDa, and 250 kDa when the assay works. Good luck!
