l am a little confused, in some papers, some researchers use toluidine blue and basic fuchsin to stain tissues, and others use traditional method such as Hematoxylin eosin staining, actually the two methods are different in staining tissues colour, but l want to know which condition to use toluidine blue or Hematoxylin eosin staining?
H&E is the gold standard for processed tissue (formalin-fixed, paraffin embedded, frozen tissue) and what pathologist typically use to screen all samples submitted to a pathology lab, it is the most common "stain" performed. Toludine blue is considered (in pathology) more of a special stain with direct application to highlight specific components (mucins, cartilage) or cell types (nerve, glial) as well as used as a "vital" stain, in vivo (oral mucosa)
Dear Min Yue , would you mind to define your preparation protocol / techniques you have in mind or are performing practically.
e.g. Histology in general on
- animal/human tissue fixed chemically/cryoprep=frozen tissue/
- processed as usual via dehydration into paraffin / resin? (watersoluble / hydrophobic);
- FFPE sectioning / cryosectioning and, respectively,
- staining then as the "crown"...

Both kind of stainings are the first choice to get an overview of the tissue situation. As Luis has mentioned H&E is an stining with is use in Antomie and Histopathology. Toluidine blue together with Thionine, Methylenblue, and Cresylviolet (so called Nissl's stain) are basic dyes . This dyes are used to stain the Nissl-substance of neurons in brain and spinal cord tissue.
