We ard injecting an expensive chemical to pre frontal cortex of rat brain via sterotaxis guided by coordinates from brain atlas. We need to check if the injection was successful by a trial with cheap dye or stain, and examining sections under the microscope. What do you suggest? What dye, is india ink a good example?
try Hoechst33342. It is fluorescent and actively taken up by cells. Visible with any epifluorescent microscope (405nm laser) within 30mins of injection. Also is very bright, stable for years at 4 degrees C and required in very low dilutions (1:10000 is plenty). Hope it helps.
