I have a confusion in calculating the amount. I took 100mg of dried sample and dissolved it in 100ml of solvents like methanol/chloroform etc. I filtered the solvent after 48 hours of incubation and then evaporated the solvent in a rotary evaporated. Then the dissolved entities in the solvent were scraped and dissolved in 2ml of solvent. When I am calculating the amount of the respective contents from the standard curve of gallic acid/ tannic acid I need to provide in the value of amount of the plant extract in gm in the equation C=c1XV/N [c=Total content of the phenol/flavonoid in mg/g; C1=the conc. of gallic/tannic acid established frm calibration curve in mg/ml; V=the volume of the extract in ml and N=the wt of the plant extract in gm]. What will be this volume of extract in ml? 100ml or 2ml?
