I am doing research on plant leave extract and calculate the total phenolic and flavonoid in it, I have got the result which is the concentration of the compounds in ppm. If I want to make it equivalent with the standard that I used, so how is the calculation? Because, based on some literatures that I reviewed, there's no exact formula, it is just calculated by multiplying the concentration of compounds with the sample volume and then divided by sample's weight. To calculate total phenolic or flavonoid content in standard equivalents, multiply the compound concentration (mg/ml) obtained from the calibration curve by the extract solution volume (ml), then divide by the sample weight (g). This gives the content as mg of standard equivalent per gram of extract (e.g., mg GAE/g or mg QE/g).
Abdelhak Maghchiche thank you so much for your answer. I want to make sure, that I do not have to include the dilution factor in this calculation, right? Because, in some literatures, it's mentioned that we have to multiply the sample concentration by the volume and also by the dilution factor, then divide it by the sample weight.
Total phenolic and total flavonoids determination depends on methods. Spectrophotometrically. we must prepare standard curve by tannic or vanillic acid when you determine total phenols and use rutin, catechin or quercetin when determine flavonoids
