Hi, I am going to add SiRNA with DOTAP liposome in different N/P (Quatenary nitrogen of DOTAP/ Phosphate of SiRNA) ratio. For instances, I want to make N/P ratio of 4/1 and I want to add SiRNA to 10µg of DOTAP. So, according to N/P= 4, how much SiRNA I need to add?
Actually, I need a clear understanding of how to calculate N/P ratio.

Dear Mr. Kahn,
you have to calculate the amount of protonable amines in your formulation, which is 1 mole N per mole DOTAP, and the amount of phosphate groups in your siRNA, which depends on the lenght of your siRNA.
