Hi everyone I am doing PAL assay in cucumber leaves and I am using the paper below as a reference
I don't understand how they convert the absorbance value to enzymatic units
They indicate that they measured the increase in OD290 over a 1 hour period and then they reported as a U/g FW
They add this info:
One unit of PAL activity was defined as the amount of enzyme which caused an increase in absorbance of 0.01 h−1

The authors are using an arbitrary unit definition: 1 unit is the amount of enzyme that causes an absorbance change of 0.01 per hour, under a specified set of conditions.
They divide the number of enzyme units (delta A290/h/100) by the amount of plant matter used to prepare the sample that was measured in grams fresh weight, accounting for dilution of the extract. So, for example, if they ground up 100 grams of plant matter in a total volume of 10 ml (10 g/ml), then used 1 ml of that in the assay, the reaction contained the equivalent of 10 grams FW. Divide the enzyme units measured by 10 g FW.
